چکیده

L’obCsitC, pathologie frtquente dans les sociCtCs industrialiskes, est caractQiste par un sur-diveloppement du tissu adipeux. Nous avons CtudiC un moditle animal d’obksitk, le rat Zucker gCnCtiquement obkse. D b l’iige de 30 jours, les cellules adipeuses des rats mutants transportent beaucoup plus de glucose que celles des temoins normaux des memes portCes. Cette anomalie repose sur une augmentation du nombre des transporteurs de glucose et plus particulihement sur une surexpression de l’isoforme GLUT4. La rtgulation de l’expression de GLUT4 par le gknotype (fa) est sptcifique du tissu adipeux. De plus, nous avons montrC recemment qu’elle passe par une suractivation du promoteur. La surexpression de GLUT4 permet une entrCe massive de glucose dans les cellules adipeuses, favorisant une lipogCn2se excessive. UltCrieurement, les niveaux adipocytaires de GLUT4 diminuent, en conjonction avec l’apparition d’une insulinoresistance. Chez l’homme obkse, la diminution de I’expression de GLUT4 dans le tissu adipeux est aggravte par le diabkte non-insulin0 dependant (DNID), un dtfaut qui concerne Cgalement une autre isoforme GLUTS, transporteur de fructose. L’implication de GLUT5 dans l’utilisation des hexoses par la cellule adipeuse et son r81e dans le DNID restent B determiner.
